The Pharmacokinetics, Pharmacodynamics and Drug Metabolism (PPDM) department of our company's Research Laboratories in West Point, PA is seeking a Senior Scientist (R3 level) to support our efforts in understanding the biotransformation and disposition of small molecule drug candidates with an emphasis on metabolite identification. PPDM continues to facilitate the discovery and development of a wide range of modalities by characterizing and understanding their in vitro and in vivo drug metabolism and disposition to help design, optimize and prioritize molecules for clinical success. The candidate will play a critical role in supporting the metabolite identification of various small molecule drug candidates from both in vitro and in vivo sources by utilizing state-of-the-art isolation equipment, Liquid Chromatography High-Resolution Mass Spectrometry (LC-HRMS) technologies and software tools.

The ideal candidate will possess the necessary skills devoted to the isolation and subsequent characterization of metabolites with a focus on metabolite identification and structure elucidation using various HRMS techniques. The candidate will isolate or generate metabolites from various matrices, isolation using UV/VIS based, mass-directed, or other fractionation techniques, and further elucidate the structure with our structural chemistry colleagues. The candidate will be part of a collaborative cross disciplinary team working closely with other research functions in West Point and sites across the global company organization (including additional PPDM groups, Medicinal Chemistry, Biology, Pharmacology and Toxicology). Effective interpretation of results and communication across disciplines will be critical to the success of the position.

Qualifications

Minimum Educational and Experience

  • PhD + ≥1 yr. experience in Drug Metabolism and Pharmacokinetics, Organic Chemistry, Pharmacology, Analytical Chemistry, Biochemistry, or relevant industry experience in the field of drug metabolism, or masters +3 years’ experience, or bachelor’s +5 years’ experience.

Job Duties

  • Isolation and characterization of unknown metabolites formed from in vivo and in vitro sources

  • Responsible for the metabolite profiling, structure elucidation and understanding of metabolic liabilities of small molecule therapeutics to support both discovery and development programs.

  • Skilled in metabolite purification and isolation (e.g. via fraction collection) from various in vitro and in vivo matrices.

  • Use of LC-HMRS systems, such as ion mobility mass spectrometry, to analyze complex metabolite mixtures

  • Design and implement new analytical capabilities and/or incorporate new technologies to improve the identification of unknown metabolites.

  • Interpretation of complex data sets, report documentation and proper communication of results with various stakeholders across the organization

Required Experience and Skills

  • In depth knowledge of drug metabolism and pharmacokinetics

  • Strong organic chemistry background

  • Experience in sample preparation techniques of xenobiotics from various biomatrices such as microsomes, hepatocytes, bile, urine, feces and plasma

  • Demonstrated expertise in metabolite identification using Liquid Chromatography High-Resolution Mass Spectrometry

  • Experience using various in vitro metabolic platforms such as liver microsomes, hepatocytes, co-culture systems, etc.

  • Solid phase extraction to semi-purify metabolites prior to subsequent purification

  • Knowledge and mechanistic understanding of drug metabolizing enzymes

  • Strong collaboration skills with proven experience in complex scientific problem solving

  • Clear communication of complex results in the context of programs being supported

  • Attention to detail, critical thinking and growth mindset

Preferred Experience and Skills

  • Understanding of Medicinal Chemistry approaches and physicochemical properties as related to drug optimization

  • Experience in using software-guided approaches for small molecule metabolite elucidation

  • Understanding and experience of ADME and PK/PD concepts in drug discovery

  • Established track record of scientific achievement through publications and presentation at scientific conferences

  • Experience in mechanisms for Phase I and Phase II metabolite formation with an understanding of organic chemistry to drive small molecule drug optimization efforts

  • Experience running various reactive metabolite trapping assays

  • Knowledge and experience with emerging analytical technologies and platforms
